ABSTRACT
Carbohydrates are a ubiquitous class of natural products that have a profound influence on a variety of physiological processes and are key components to the activity of numerous natural products such as erythromycin. In general, protecting groups play a critical role in oligosaccharide synthesis, as well as in oligonucleotides such as DNA, and RNA synthesis. The careful selection of the protecting group family and the synthesis route are intertwined and can impact the quality and desired attributes of the final API such as yield, purity, RSMs and COG.
In this talk, two topics will be covered:
How various protecting groups influence the rate and stereochemical outcome of glycosylation through reactivity tuning, hydrogen bonding, neighboring group participation, stereoelectronic effects and conformational control.
Protecting groups not only affect glycosylation but have a profound effect on numerous other chemical reactions. Some examples from synthetic processes that are not related to the process of glycosylation such as the metathesis reaction, additions to imines and carbonyls, among other reactions, will be presented. Overall, this is a very broad subject that can only be briefly covered in this presentation, but the idea is to present the fact that protecting groups are generally not innocent bystanders that can be ignored.
