ABSTRACT
The number of carbohydrate-based and cyclic peptides drugs have been increasing and several were recently approved or are in clinical trials. In general, the use of protecting groups for their synthesis at small scale and industrial manufacturing is ubiquitous. The careful selection of the protecting groups and the synthesis route are intertwined and can impact the quality and desired attributes in the final API such as yield, purity, RSMs and COG.
In this talk, two topics will be covered:
- Strategies for late-functionalization of a cyclic peptide antibiotic (Aureobasidin) preempting the need to use protected unnatural aminoacids (SPPS) and cyclization to make a chemically diverse series of analogs.
- The effect of protecting groups on chemical glycosylation by changing the reactivity, neighbouring group effect and conformational changes in carbohydrates and related compounds.
BRIEF BIO
Dr. Peter G. M. Wuts is globally recognized as an expert in organic synthesis. His expertise spans process development, methodology development, and total synthesis. Throughout his distinguished career at Upjohn, Pharmacia & Upjohn, Pharmacia, Pfizer, and Kalexsyn, Dr. Wuts has made invaluable scientific contributions. He has extensive experience in synthesizing steroids, aminocyclitol antibiotics, peptide isosteres, polycyclic aromatic hydrocarbons, paclitaxel, cyclic peptides, and other heterocycles. Dr. Wuts has authored over 90 scientific publications and is also the author of “Protective Groups in Organic Synthesis”
